A comprehensive review in Cell Press Blue analysed over 350 studies on protein and amino acid restriction across species. Restricting total dietary protein — or specifically methionine, isoleucine, and valine — extended lifespan and improved metabolic markers in multiple animal models; one mouse study showed a 23% increase in male lifespan through valine restriction alone. The authors note human evidence remains limited and protein needs vary considerably with age and activity level.

XPRIZE Healthspan advances 20 therapeutic teams into formal clinical trials

XPRIZE announced that 20 finalist teams from its $101 million Healthspan competition have entered formal clinical trials testing whether therapeutics can restore muscle, cognitive, and immune function in adults aged 50-80. Ten teams received $1 million Milestone 2 awards. Trials run through 2029, coordinated through the University of Utah Data Coordinating Centre. DNA mutation accumulation sets a hard ceiling on human lifespan near 194 years

A study modelling intrinsic somatic mutation rates across tissues found that even under the most favourable biological and environmental conditions, the rate at which cells accumulate DNA copying errors imposes a theoretical ceiling on human lifespan in the range of 146 to 194 years — a constraint not addressable by current anti-senescence, telomere, or inflammation-targeting interventions.

Why it mattersPositions radical life extension as a DNA replication-fidelity problem specifically, rather than an inflammation, senescence, or metabolic problem. Most longevity drug pipelines are not targeting that layer.
