Nature Aging: TNFR1 links chronic inflammation to intestinal stem cell ageing via fatty acid oxidation

A Nature Aging study finds that TNFR1 signalling — the main inflammatory receptor pathway — directly impairs fatty acid oxidation in intestinal stem cells, reducing their regenerative capacity. The effect accumulates with age and is reversible in animal models when the pathway is blocked.

Why it mattersIntestinal stem cell renewal underpins gut barrier function and immune homeostasis. A single, targetable receptor linking inflammation to this decline is a cleaner mechanistic handle than most ageing pathways the field has proposed.

Nature Aging: Maladaptive trained immunity in haematopoietic stem cells drives inflammageing

A second Nature Aging study published this week identifies maladaptive trained immunity in haematopoietic stem cells as a driver of inflammageing — the low-grade chronic inflammation associated with biological ageing. The finding links immune memory in bone marrow stem cells directly to systemic ageing phenotypes.

Why it mattersTrained immunity is typically studied as a protective mechanism. Identifying its maladaptive form as an upstream driver of inflammageing connects haematopoietic biology to the broader ageing field in a way that opens new intervention targets.

Lancet study: stopping statins in adults over 75 at low cardiovascular risk did not increase deaths

A randomised Lancet trial found that discontinuing statin therapy in adults aged 75 or older assessed as low cardiovascular risk did not result in increased mortality or cardiovascular events over the follow-up period. The study compared discontinuation to continuation in patients not previously managed for established cardiovascular disease.

Why it mattersStatin deprescribing in older adults has been resisted partly because the evidence for stopping was weak. This trial provides the first direct randomised data on the question for a specific older-adult risk group.

Social isolation harms the ageing brain independently of loneliness, study finds

A new study finds that objective social isolation — measured by frequency of social contact — is associated with accelerated brain ageing and cognitive decline in older adults independently of subjective loneliness. The two constructs produce distinct neurobiological signatures on imaging.

Why it mattersLoneliness and isolation are frequently conflated in public health discussion. Separating their brain effects means they may require different interventions — and one can cause damage without the other being present.

Framingham data: accelerated biological age in midlife predicts dementia and cardiovascular disease decades later

A Scientific Reports analysis of nearly three decades of Framingham Offspring Study data found that accelerated biological age — measured in midlife — is associated with significantly elevated risk of cardiovascular disease, dementia, and all-cause mortality, with the dementia signal strongest in women.

Why it mattersIf biological age measured in midlife predicts dementia risk with decades of lead time, it becomes a plausible target for early screening. The Framingham cohort is the most longitudinally robust cardiovascular dataset in existence.
